背景介紹：DHRS11 is a 260 amino acid secreted protein. Belonging to the short-chain dehydrogenases/reductases (SDR) family, DHRS11 forms a homotetramer and is expressed as two isoforms produced by alternative splicing events. The gene that encodes DHRS11 maps to human chromosome 17, which makes up over 2.5% of the human genome with about 81 million bases encoding over 1,200 genes. Two key tumor suppressor genes are associated with chromosome 17, namely, p53 and BRCA1. Tumor suppressor p53 is necessary for maintenance of cellular genetic integrity by moderating cell fate through DNA repair versus cell death. Malfunction or loss of p53 expression is associated with malignant cell growth and Li-Fraumeni syndrome. Like p53, BRCA1 is directly involved in DNA repair, though specifically it is recognized as a genetic determinant of early onset breast cancer and predisposition to cancers of the ovary, colon, prostate gland and fallopian tubes. Chromosome 17 is also linked to neurofibromatosis, a condition characterized by neural and epidermal lesions, and dysregulated Schwann cell growth. Alexander disease, Birt-Hogg-Dube syndrome and Canavan disease are also associated with chromosome 17.

流式檢測樣品的制備

提示

1、流式細胞儀檢測需要將細胞制備成單細胞懸液。

2、避免細胞成團，以免堵塞流式細胞儀。

3、實體組織制備單細胞懸液需要物理方法分離或酶消化法進行，具體組織的方法，依照經驗來進行。

方案A: 培養細胞的制備

實驗材料：0.25%胰，15ml或50ml的離心管。

實驗流程：

1、如果是懸浮細胞，則小心將細胞移入離心管中，進行計數和活力分析。進行步驟3。

2、如果是貼壁細胞，建議使用0.25%的*消化細胞，制備成單細胞懸液后置于離心管中計數和活力分析。

3、200g 離心5min。棄去上清，應用適量的流式孵育緩沖液重懸細胞，調整細胞濃度為1×107/ml。

方案B: 組織細胞制備。

實驗材料與試劑：剪刀和鑷子，載玻片， 60×15mm的組織培養皿；細胞過濾器（尼龍網過濾）,0.01MPBS。
