背景介紹：Component of the SMC5-SMC6 complex, a complex involved in repair of DNA double-strand breaks by homologous recombination. The complex may promote sister chromatid homologous recombination by recruiting the SMC1-SMC3 cohesin complex to double-strand breaks. The complex is required for omere maintenance via recombination in ALT (alternative lengthening of omeres) cell lines and mediates sumoylation of shelterin complex (osome) components which is proposed to lead to shelterin complex disassembly in ALT-associated PML bodies (APBs). In vitro enhances ubiquitin ligase activity of NSMCE1. Proposed to act through recruitment and/or stabilization of the Ubl-conjugating enzyme (E2) at the E3:substrate complex. May be a growth suppressor that facilitates the entry of the cell into cell cycle arrest.

注意事項

1.  標本處理：（1）全血實驗  避免使用絡合鈣的抗凝劑，如ACD與EDTA，因為它們會限制鈣依賴性激活過程，*用*。此外LPS，一種常見的生物試劑污染物，是強細胞激活劑，可能會混淆試驗結果。血樣在8小時內分析，超過8小時會導致活性的損失，一般細胞因子陽性細胞會減少5%。如不能在8小時之內檢測，應將真空*水平室溫放置。（2）組織樣本制備  取組織時去除組織內凝固的。（3）固定，穿膜的試劑PH一定要正確。

2.  選擇合適的對照：為保證結合的真實和可靠性，至少熒光設置同型對照：用于消除由于抗體非特異性結合到細胞表面而產生的背景染色；陰性對照(Isotype Control)：非特異熒光的強弱取決于抗體濃度、單克隆熒光抗體特異性和純度，應與試驗管抗體相對應。在多色分析時，同型對照應與其它抗體同時使用，以避免補償造成的誤差。
